The tomato (Solanum lycopersicum L.) is a major crop of global economic significance. characterization genes associated with agriculturally important traits often performed using genetic transformation. To achieve an efficient transformation protocol, three components are required, namely, regenerable target tissue, DNA delivery method, and robust transformant selection system. present study was conducted to optimize the in vitro regeneration protocol for cv. MT1. capacity hypocotyl cotyledon explants evaluated total 20 concentration combinations two plant growth regulators (PGRs) added into basal MSB5 medium, 6-benzylaminopurine (BAP) (0, 1, 2, 3, 4 mg/L) indole-3-acetic acid (IAA) 0.05, 0.1, 0.5 mg/L). optimal PGRs cotyledons hypocotyls were supplemented 2 mg/L BAP IAA 0.1 IAA, respectively. determine minimum inhibitory (MIC) kanamycin, eight different concentrations 50, 75, 100, 125, 150, 175, 200 IAA.
